The objective of the present study is to develop medium and culture methods for production of higher alcohols using syngas fermentation. Growth of C. carboxidivorans during syngas BIX 01294 in defined medium without complex nutrients is reported in the present study. Accordingly, a minimized medium was sought to promote selectivity for butanol and hexanol in a production fermenter. Fermentation was monitored through successive modifications of the defined medium to assess the potential for commercial production of butanol and hexanol from syngas. The performance of C. carboxidivorans in batch culture using minimal defined medium was highly dependent on culture technique. A program of agitation, gas supply and pH adjustment was developed to sustain timely growth of C. carboxidivorans for culture maintenance and syngas fermentation.
C. carboxidivorans strain P7 ( Liou et al., 2005) was used in all experiments. Cited costs are for laboratory grade chemicals purchased in quantities of 1 kg or less, with prices updated December 10, 2014 (Sigma–Aldrich, St. Louis, MO, or Fisher Scientific, Waltham, MA). Selected nutrient components were studied in serum bottle batch fermentation to allow replication of experiments in a timely manner.
